Course Overview:

The course aims at introducing high end technologies related to the study of molecular interactions from and in biological systems. The course will cover the whole workflow for kinetics and thermodynamics characterization of binding events, from cutting edge protein expression and purification to high sensitivity binding assays. The bridge to “in vivo” will be crossed with the introduction to advanced light microscopy techniques tostudy the dynamic interactions of proteins within the living cell.

Interactions between biomolecules play central roles in every life process. To study them a wide range of methods have been developed over the years. The course will present a multidimensional and integrated approach to the characterization of molecular interactions.

 

The course will include lectures on:

  • state of the art expression and purification methods to handle protein complexes, membrane proteins and proteins with poor solubility (directed evolution of soluble constructs and expression in Eukariotic systems).

  • surface plasmon resonance (SPR), isothermal titration calorimetry (ITC) and Microscale Thermophoresis and their application to the study of fundamental biological processes and to drug discovery.

  • fluorescence resonance energy transfer (FRET), fluorescence lifetime imaging microscopy (FLIM) and fluorescence correlation spectroscopy (FCS) applied to study protein-protein interactions and dynamics in live cells.

 

The course will include pacticals on:

  • surface plasmon resonance (SPR), isothermal titration calorimetry (ITC) 

  • fluorescence resonance energy transfer (FRET)

 

Participants can apply to work with their own samples
