Applications are invited for a postdoctoral position to join our group focusing on the characterization of spindle coupling to cortical polarity in asymmetric cell divisions. Characterization of the molecular mechanisms of mitotic spindle coupling to cortical polarityThe correct placement of the mitotic spindle during mitosis is fundamental in determining the relative position of the daughter cells, and hence in preserving tissue architecture. Furthermore, in Drosophila germline stem cells and in mouse neural stem cells the orientation of the mitotic spindle ensures that only one of the two siblings retains niche contacts and stem-like properties. Recent data support the view that the conserved NuMA/LGN/G?i pathway is responsible for anchoring astral microtubules to the correct cortical sites in diverse cellular systems, although with a different coordination to the cellular polarity. In particular, issues address have been of 1) how these force generating machines are clustered at restricted cortical sites; 2) how they are coordinated with the polarity proteins Par3/Par6/aPKC. However, regulatory circuits and ancillary subunits orchestrating the function of NuMA/LGN/G?i in living cells still remain to be identified. The general goal of this project is the structural characterization of known protein complexes involved in spindle placement, and the identification new components of the network. The strategy to achieve this aim will combine X-ray crystallography, biochemistry and cell biology. We are looking for a highly motivated and enthusiastic postdoctoral fellow who enjoys working as part of a collaborative and multidisciplinary team. The ideal candidate should have a PhD in protein structure, biochemistry or equivalent qualification, and extensive experience in protein expression and characterization. Previous experience in protein structure determination would be desirable.The Structural Biology Department of the IFOM-IEO Campus is equipped with the state-of-the-art apparatus for protein purification and crystallization, including a nanodrop crystallization robot and an automated imaging system, and has good access to the synchrotron beamlines.
