Diffraction techniques core facility provides services for characterization of protein structure using single crystal X-ray diffraction and small-angle X ray scattering for analysis of liquid samples. Both methodes are backer by advanced approaches available in laboratory. The quality of high-intense X-ray beams is ensured by using of MetalJet sources (Excillum). The provided services include assistance with data processing and reporting ready for publication.

Instruments Available:

Preparation of biomolecule crystal samples for the measurement in the diffractometer. Handling of crystals, testing of cryo-conditions, cryo-cooling of crystalline samples.

Processing of X-ray diffraction data. Upon request, assistance with structure determination is also offered.

High performanece in-lab diffractometer for single crystal diffraction experiments. The instrument hosts MetalJet D2 X-ray source with Ga anode (λ=1.34Å) and large Photon III detector. The instrument is capable of both screening and data collection, including experimental phasing experiments.

Additional equipment:

  • CryoStream 800 - cooling for for experiments at cryogenic temperatures
  • ISXstage - motorized holder for in-situ testing of crystal quality in the crystallization plates. Limited data collection is also possible.
  • HC-Lab (Arinax) - humidity control of the crystal environment at the room temperature. Used for crystal dehydration experiments, which can improve the crystal quality

In-situ diffraction in crysttalization plates

The versatile SAXSpoint 2.0 (Anton Paar) SAXS/WAXS device is equipped with the latest high brilliance C2+ liquid MetalJet source (liquid gallium alloy anode) and a Dectris EIGER 1M detector. Liquid samples are loaded automatically using the automated high performance sampler or manually in flow through measurement cells, including a high S/N silicon nitride measurement cell. Measurements are performed in the temperature range -10 °C to 120 °C. The state of the sample can be monitored online using in situ UV-Vis spectroscopy to identify radiation damage, and measure precise sample concentration and aggregation state.
